We just did gram staining for Sean since his bio was contaminated we do not know what it is but we guessed it’s streptococcus or diplococci gram positive and the rest of the day we just cleaned up a lot of slides

This week we where trying to do dna isolation again but our broth was actually contaminated and we had to wait for more kits to finish the rest of the isolation we did the previous weeks.  DNA isolation  is a process of purification of  DNA  from sample using a combination of physical and chemical methods.

last week we just did blasting on ncbi and learned more about PCR. The  program compares nucleotide or protein sequences to sequence databases and calculates the statistical significance of matches.  BLAST  can be  used to  infer functional and evolutionary relationships between sequences as well as help identify members of gene families.

This week we did DNA isolation with Chad which is a process of purification of  DNA  from sample using a combination of physical and chemical methods. This day felt like I was doing my own experiment I did it with a partner and asked questions when I needed to. But using the micropipets by myself and learning about new machines and techniques was very exciting. Especially this was a very long, easy but fun experiment to do! I’m excited to do what’s next

Last week we helped Stacy with 24 slides to gram stain which was very time consuming but very fun! We did not know what they were she was the only one that knew but here are some slides I collected. Circle and rods either gram positive or negative. Also we set things up for chad paracallic pump which we just set some tubings, spacers, bio films for it to be autoclave and to be set up after.

Gel electrophoresis is a method for separation and analysis of macromolecules and their fragments, based on their size and charge. We ended up making a gel and also when putting them in the electrophoresis which 123 where the control, 4 being the GCC Taq, 5 and 5 being PTAK which wanting Apx. 800Bp. This method is to identify the piece of dna 🧬 and certain size and conform that DNA. Larger sizes don’t go father then smaller sizes.
